ABSTRACT

ObjectiveTo investigate the effects of different acupuncture schemes on behaviors, neurotransmitters and inflammation-related factors in post-stroke depressed (PSD) rats. MethodsA total of 72 healthy male Sprague-Dawley rats were randomly divided into normal group, model group, drug control group, scalp acupuncture group, abdominal acupuncture group and combined acupuncture group, 12 rats in each group. The PSD model was prepared using a combination of middle cerebral artery occlusion and chronic unpredictable mild stimulation (CUMS) for the model and each intervention groups. The drug control group was administered fluoxetine, the scalp acupuncture group accepted acupuncture at Baihui (DU 20) and Yintang (EX-HN3), while the abdominal acupuncture group at Zhongwan (RN 12) and Guanyuan (RN 4), the combined acupuncture group at all the four acupoints, for 21 days. They were assessed with Longa neurological function score, body mass, open-field test and sugar-water preference test on the 0 (before modeling), the seventh (before CUMS), the 14th (before treatment) and the 35th day (after treatment). The levels of 5-hydroxytryptamine (5-HT), dopamine (DA), norepinephrine (NE), interferon-γ (INF-γ), tumor necrosis factor⁃α (TNF⁃α) and macrophage migration inhibitory factor (MIF) in the serum were determined with ELISA on the 35th day. Results The neurological function score was lower in the drug control group and the combined acupuncture group thanin the model group (P < 0.01) on the 35th day; while the body mass was higher in the drug control group, the abdominal acupuncture group and the combined acupuncture group than in the model group (P < 0.01); and the number of horizontal span frames, the number of uprightness and the sugar water consumption were higher in the drug control group, the scalp acupuncture group, the abdominal acupuncture group and the combined acupuncture group than in the model group (P < 0.05). There was no significant difference among the four intervention groups (P > 0.05). The levels of 5-HT, DA and NE were higher in the four intervention groups than in the model group (P < 0.01), and the levels of IFN-γ, TNF⁃α and MIF were lower (P < 0.01). 5-HT level was higher in the combined acupuncture group than in the scalp acupuncture group and the abdominal acupuncture group, and the levels of IFN-γ, TNF⁃α and MIF were lower (P < 0.01); the levels of DA and NE was higher in the combined acupuncture group than in the abdominal acupuncture group (P < 0.01). ConclusionThe combination of scalp acupuncture and abdominal acupuncture can improve behavior, neurotransmitters and inflammatory factors of PSD rats, like fluoxetine. Scalp or abdominal acupuncture alone is still effecive, and can be used secondarily.

ABSTRACT

BACKGROUND: The degradation speed of biological materials is critical for the clinical use of guided bone regeneration. The partial biological characteristics and treatment efficacy of concentrate growth factor (CGF) fibrin have been explored preliminarily, but its degradation properties have not yet been reported.OBJECTIVE: To explore the degradation properties of CGF fibrin and platelet-rich fibrin (PRF) in artificial saliva and compare the degradation speed of these two biological products.METHODS: Ten volunteers were selected, and 18 mL of venous blood from each volunteer was extracted and stored in two vacuum blood collectors. The blood samples were then placed into the drum of the Medifuge centrifugal acceleration machine, to separate CGF fibrin and PRF specimens following the preparation process, respectively. Both CGF fibrin and PRF specimens were respectively made into bulk and membranoid, and were then immersed in artificial saliva under 37 ℃. The mass of the bulk specimens and area of the membranoid specimens were measured regularly, and the degradation processes of CGF and PRF were recorded. The degradation curves were drawn to compare the degradation speed of CGF fibrin and PRF.RESULTS AND CONCLUSION: The mass of CGF fibrin and PRF showed no significant difference at the 5th day (P >0.05), while the mass of CGF fibrin was higher than that of PRF at the 3rd, 4th and 6th days (P < 0.05). The residual area of CRF was significantly larger than that of PRF at posttreatment 1-6 days (P < 0.05). To conclude, the degradation speed of bulk or membranoid CRF is slow than that of PRF in artificial saliva. The higher the fibrin content is, the slower the degradation ability is, indicating the strong bioreproductive function.

ABSTRACT

Objective To detect the independently designed synthetic peptide adsorbed to the titanium surface and its inhibitory effect on streptococcus gordonii, and to provide a new means for antibiosis reseach on oral implants. Methods The physical and chemical properties of the synthetic peptide and antimicrobial peptide were measured by ExPASy Prot?Param tool, ProtScale analysis, circular dichroism and Zeta potential instrument. The synthetic peptide was anchored on the surface of the titanium specimen through incubation at room temperature. The adsorption of the synthetic peptide to the titani?um surface was examined by X-ray photoelectron spectroscopy (XPS) and the atomic force microscope (AFM). The inhibitory effect on streptococcus gordonii of the synthetic peptide fixed on the titanium surface was viewed by confocal laser scanning microscopy (CLSM). The destructive effects of the synthetic peptide and the antimicrobial peptide on streptococcus gordonii were observed through the transmission electron microscope (TEM). Results The independently designed synthetic peptide still had the physical and chemical properties that the antimicrobial peptide desired. The synthetic peptide had already been detected on the titanium surface after incubated in a 5 g/L synthetic peptide solution. The titanium specimen fixed with the synthetic peptide inhibited the survival and adhesion of streptococcus gordonii. Conclusion It suggests that the indepen?dently designed synthetic peptide might have reached the goal of bacterial inhibition on the titanium surface.

ABSTRACT

BACKGROUND:With the development of the oral cavity planting technology, dental implants have been strongly retained and stabilized, characterized as both aesthetic appearance and functional achievements. Especial y, they are accepted by more and more patients due to no damage to adjacent teeth. However, diabetes may not only lead to periodontal disease, but also result in delayed healing of the oral mucosa and alveolar bone as wel as infection. OBJECTIVE:To summarize the relationship between diabetes and peri-implantitis, and focus on the maintenance of dental implants for diabetic patients. METHODS:A computer retrieval of PubMed and CNKI databases was performed for relevant articles published from January 1980 to January 2016 using the keywords of“diabetes;peri-implantitis”in English and Chinese, respectively. RESULTS AND CONCLUSION:We confirm that diabetes is a relative contraindication to implant restoration, but the use of a series of methods that ensure the successful osseointegration, including blood glucose control, strictly prohibiting early loading or multi-implant restoration for single alveolar bone, anti-infection, implant surface modification, can make high success rate for diabetic patients. This brings the gospel to diabetic patients who hope to solve the problem of missing teeth by implant-supported prosthesis.

ABSTRACT

Objective To investigate the remineralizing therapy of deep caries and in vitro biomimetic remineralization of demineralized dentin by phosphorylated chitosan/amorphous calcium phosphate compound (P-chi/ACP) and tripolyphosphate (TPP). Methods Thirty-two extracted human molars were cut and completely demineralized. Two samples were used to show the characteristics by transmission electron microscopy (TEM). The other 30 samples were divided into two groups:fifteen samples were treated by P-chi/ACP and TPP (P-chi/ACP+TPP group), the other fifteen samples were not treated by TPP (P-chi/ACP group). All of the samples were distinguished into experimental side and control side, and then they were set on the in vitro model for 1 week. Micro-computed tomography (μCT) and TEM were used to assess the effects of remineralization. Results μCT detection revealed that the mineral density were higher in the experimental sides (125.42±12.16 and 119.39±8.64) than that of control sides (96.96±10.56 and 105.27±9.42) in both groups (P<0.01). TEM figures showed that hierarchical intrafibrillar remineralization was realized in samples of P-chi/ACP+TPP group, while trace amounts of hierarchical remineralization was detected in P-chi/ACP group. Conclusion Fully demineralized dentin appears to have the potential to be remineralized with the application of P-chi/ACP. The ultrastructure of samples is better in P-chi/ACP+TPP group than that of P-chi/ACP group.

ABSTRACT

Objective To detect the inhibitory effect of siliver nanoparticles loaded titanium nanotubes on staphylococcus aureus, and provide a theoretical basis for implant local application. Methods Orderly arrangement of titania nanotubes produced by anodic oxidation, loaded silver nanoparticals by situ replacement. Scanning electron microscopy (SEM) and transmission electron microscopy(TEM) were used to detect the morphology topology of silver nanoparticals, titanium nanotubes and siliver particals loaded titanium nanotubes. The minimum inhibitory concentration of silver nanoparticles was calculated. The antibacterial of planktonic bacteria was detected 1 day, 3 days and 5 days after culturing staphylococcus aureus on siliver particals loaded titanium nanotubes. The inhibitory bacterial adhesion properties were detected by scanning electron microscopy. Results The uniform and orderly diameter of 80~120 nm TiO2 nanotubes were prepared under 18 V voltage, loaded diameter of 20 nm silver nanoparticals, which effectively inhibited adhesion and proliferation of staphylococcus aureus. Conclusion Titanium nanotubes produced by 18 V have a stronger drug loading capacity. The 100 mmol/L silver nanopartical solution loaded nanotubes can effectively inhibit staphylococcus aureus adhesion and proliferation within three days.

ABSTRACT

Objective To explore the effects of electroacupuncture in different time on the variations of fractional anisotropy (FA) mean value in diffusion tensor tractography (DTT) in spinal cord injured (SCI) rats. Methods 48 Sprague-Dawley rats were performed with Al-len's method by NYU blow device, resulting in T10-11 SCI. They were divided into groups according to random table. Control group (n=16) re-ceived no treatment. Experimental group (Group A, n=32) received electric stimulation in the scalp surface projection area of motor area and local electric stimulation at damaged site. Group A was divided into Group A1 (n=16) and Group A2 (n=16) again. Group A1 received elec-tric stimulation 3 days after SCI, while Group A2 received electric stimulation 2 weeks after SCI. They were assessed with Basso-Beattie-Bresnahan (BBB) scores and routine MRI and diffusion tensor imaging (DTI). Results There was no significant difference in BBB score among all the groups at the first week (P>0.05). BBB scores of the Groups A1 and A2 were significantly different from the control group at the second and fourth week (P0.05). There was significant difference among the Groups A1, A2 and the control group at the fourth week (P<0.001). Group A1 was better than Group A2 (P<0.001). DTT images of all the groups were successful recon-structed, the Groups A1 and A2 repaired better than the control group. Conclusion The earlier electroacupuncture is taken after spinal cord injury, the better the outcome is.

Methods In this study the subjects adopted are clean grade rats, which were performed with Allen's method by NYU blow device, resulting in SCI models. 192 rats successfully modeled were divided into groups according to the random table. Group A received electric stimulation in the scalp surface projection area of motor area: Group A1 received electric stimulation and transplantation of umbilical cord blood stem cell, while Group A2 only received electric stimulation; Group B received local electric stimulation at damaged site: Group B1 received electric stimulation and transplantation of umbilical cord blood stem cell, while Group B2 only received electric stimulation; Group C received electric stimulation in the scalp surface projection area of motor area and local electric stimulation at damaged site: Group C1 received electric stimulation and transplantation of umbilical cord blood stem cell, while Group C2 only received electric stimulation; Group D are just the SCI models: Group D1 received transplantation of umbilical cord blood stem cell without electric stimulation, while Group D2 neither received cell transplantation nor electric stimulation. The sacrifice time of rats was the 1st week, 2nd week, 3rd week, 4th week, 8th week, and 12th week after operation respectively. The distribution of the remained or regenerative neurons at damaged areas by corticospinal tract anterograde tracer with Biotinylated dextran amine (BDA), and we also detected the content changes of GAP-43, MBP and the MAB1281 specific expression of the human nucleus of human umbilical cord blood stem cells. Results 1). NYU is a blow device for Allen's rat model of SCI, which could quantify the crash potential energy, and gain a stable and repeated model with high successful rate. 2). At each time site in this study, the factors that benefit for the neurofunction restoration after electric stimulation in the microenvironment into which the stem cells have been transplanted are GAP-43 and MBP, and they are all positive; the effects of the combination of head acupuncture with body acupuncture are much better than single head acupuncture or body acupuncture. 3). At each time site in this study, the factors that benefit for the neurofunction restoration after injury are GAP-43 and MBP, and the effects of them are all positive, which do not depend on whether the stem cell transplantation has been done. Conclusion 1). After the stem cell transplantation have been done, the changes of the microenvironment develops in the direction of benefiting for restoring neurofunction, which means that stem cell transplantation can promote the restoration of neurofunction and the stem cell transplantation is successful in this study. 2). Electric stimulation is significantly positive for the factors (GAP-43, MBP) which benefit for restoring neurofunction in the microenvironment after stem cell transplantation, the effects of the combination of head acupuncture with body acupuncture is better than single head acupuncture or body acupuncture. The significantly positive effects of the electric stimulation on microenvironment after injuries are independent, which do not depend on whether the stem cell transplantation has been done. Electric stimulation and stem cell transplantation are significantly synergistic at the microenvironment.
